A separate phase 2 trial in type 2 diabetes reported meaningful reductions in HbA1c and body weight 3. In a substudy focused on liver fat, a majority of participants in the higher-dose groups reached liver-fat levels within the normal range by 48 weeks 4.

Tri-agonism does not just add a third pathway. It changes the balance between eating less and spending more.

Safety signals observed

As with other incretin-based compounds, the most frequent adverse events in trials were gastrointestinal (nausea, diarrhea, vomiting), largely mild to moderate and more common during dose escalation. Dose-related increases in heart rate were also reported 2. These observations come from controlled clinical settings with medical supervision.

• How much of the weight lost is fat mass versus lean mass, and how that compares across agonist classes.

Growth hormone (GH) is not secreted at a steady rate. The pituitary releases it in bursts, most prominently during deep sleep, and this pulsatile pattern is part of how tissues read the signal. The three regulators of the GH axis

CJC-1295 and the DAC modification

Native GHRH and Sermorelin are cleared within minutes. CJC-1295 with DAC (Drug Affinity Complex) carries a reactive group that binds covalently to circulating albumin, dramatically extending its presence in the body. Tesamorelin and visceral fat

1. Start with a falsifiable question

"Does peptide X help recovery?" is not testable. "Does peptide X increase fibroblast migration in a scratch assay compared with vehicle at 24 hours?" is. A precise question defines the model, the comparison, the endpoint and the time point all at once.

2. Choose the right model

Cell lines are fast and controllable but simplified. Primary cells and tissue explants add realism at the cost of variability. Animal models capture whole-organism effects and come with ethical obligations and regulatory oversight. Pick the simplest model that can actually answer your question.

3. Define controls

• Vehicle control: the same solvent, volume and handling, without the peptide.
• Positive control: a compound with a known effect, proving the assay can detect one.
• Single-component controls when studying blends, to see which ingredient drives the result.

4. Randomize and blind

Assign samples or subjects to groups randomly, and keep the person measuring outcomes unaware of group allocation. These two steps remove the most common sources of unconscious bias, and they are among the items most often missing from published studies 1.

5. Decide the sample size in advance

Underpowered studies produce noisy results that are hard to interpret and easy to over-read. Use a power calculation based on the effect size you expect and the variability of your assay, and state the number before you start.

Every research peptide is sold with a number attached to it, usually something like ≥98% or 99%. That number is only meaningful if you know how it was produced. A purity figure is the output of an analytical method run on one specific batch, on one specific date, by one specific laboratory. Change any of those and the number can change too.

Purity is a measurement, not a label

Synthetic peptides are built one amino acid at a time, usually by solid-phase peptide synthesis. Each coupling step is highly efficient but never perfect, so every crude batch contains small amounts of closely related molecules: sequences missing one residue (deletion peptides), truncated chains, incompletely deprotected side chains, oxidized or deamidated variants 2. Purification removes most of them. The purity figure tells you how much remains.

That is why a reputable supplier certifies each batch individually. Two vials of the same compound from different syntheses are, analytically speaking, two different samples.

What HPLC measures

High-performance liquid chromatography (HPLC), almost always in reverse-phase mode, separates the components of a sample by how strongly they interact with a hydrophobic column. As the sample travels through, each component exits at a characteristic retention time and is detected by UV absorbance, typically around 214–220 nm, where the peptide bond absorbs strongly 1.

The result is a chromatogram: a trace with one dominant peak (the target peptide) and, ideally, only small peaks around it. Purity is reported as area percent: the area of the main peak divided by the total area of all detected peaks.

What mass spectrometry adds

A sample can be 99% pure and still be the wrong compound. Mass spectrometry (MS) measures the molecular weight of what is in the main peak and compares it with the theoretical mass of the intended sequence. When the observed and theoretical masses match within the instrument's tolerance, identity is confirmed. HPLC answers how much; MS answers what.

How to read a COA in five checks

• Batch or lot number that matches the number printed on the vial.
• Method stated explicitly (e.g. RP-HPLC with UV detection; ESI-MS for identity).
• Chromatogram attached, not just a number typed into a table.
• Observed vs. theoretical mass reported side by side.
• Laboratory name and test date, ideally an independent third party.

Why the number changes from batch to batch

Small variations between syntheses, purification runs and lyophilization cycles are normal. A batch certified at 99.2% and the next at 98.9% is not a red flag. What would be a red flag is a COA that never changes, carries no batch number, or cannot be traced to a laboratory. Identical numbers across many batches usually mean the document is generic.

A purity number without a batch number is a marketing claim. With a batch number, a method and a chromatogram, it becomes evidence.

Three questions to ask before you trust a formulation

• Does this COA correspond to the exact batch I am receiving?
• Were both purity (HPLC) and identity (MS) tested?
• Who performed the analysis, and can I see the raw data?

Why peptides degrade in solution

Once dissolved, peptides are exposed to several well-described degradation pathways 1:

• Oxidation of methionine, cysteine and tryptophan residues, accelerated by light and dissolved oxygen.
• Deamidation of asparagine and glutamine, especially in Asn-Gly sequences, accelerated by heat and higher pH.
• Aggregation, often triggered by agitation and air–liquid interfaces.
• Adsorption to glass and plastic surfaces at very low concentrations.

Choosing the solvent

For most research peptides, the standard choices are sterile water or bacteriostatic water (sterile water with 0.9% benzyl alcohol as a preservative) 2. Bacteriostatic water is preferred when a vial will be accessed more than once, because the preservative limits microbial growth between uses. A few hydrophobic sequences require small amounts of other solvents; the supplier's technical data should guide that choice.

Step-by-step reconstitution

• Let the sealed vial reach room temperature before opening, to avoid condensation inside.
• Wipe the rubber septum of both vials with 70% isopropyl alcohol and let it dry.
• Draw the solvent with a sterile syringe and inject it slowly down the inner wall of the vial, not directly onto the powder.
• Swirl or roll gently until the cake dissolves. Never shake: shear and foaming promote aggregation.
• Inspect the solution. It should be clear and free of particles.
• Label the vial with compound, concentration, solvent and date.

Calculating concentration

Concentration is simply mass divided by volume. A vial containing 10 mg of peptide reconstituted with 2 mL of solvent yields a 5 mg/mL solution. Adding 1 mL instead yields 10 mg/mL. Record the value on the label and in your lab notebook so every downstream calculation in the experiment starts from the same number.

Storage and aliquoting

Before reconstitution, keep vials sealed, protected from light and moisture, refrigerated for routine use or frozen at −20 °C for long-term storage. After reconstitution, keep the solution at 2–8 °C, away from light. If a solution will be used over a long period, divide it into single-use aliquots so the main stock is not repeatedly warmed, punctured or frozen and thawed.
